Nanocarrier Based Drug Delivery: Revolutionizing Targeted Therapy

Summary
Problem with Conventional Cancer Therapy: Surgery, radiation, and chemotherapy are invasive, nonspecific, toxic, and cause severe side effects. Role of Nanotechnology: Nanoparticles (liposomes, lipid nanoparticles, micelles, dendrimers, etc.) improve drug delivery, pharmacokinetics, and therapeutic outcomes by targeting drugs directly to tumor sites. Solid Lipid Nanoparticles (SLN): Biodegradable, biocompatible, and versatile for both hydrophilic & hydrophobic drugs. Advantages: controlled release, improved bioavailability, multi-route administration, scalability, and potential for ligand-based targeting. Functionalization for Targeting: SLNs can be surface-modified with ligands (folic acid, peptides, proteins, antibodies, sugars, etc.) for active targeting to specific cells/tissues (e.g., brain, liver, tumors). Case Studies: Docetaxel + Ketoconazole SLNs - enhanced brain uptake. B-Hydroxybutyric acid grafted SLNs - improved BBB transport. Adenosine-conjugated SLNs - effective against breast/prostate cancers. Transferrin-functionalized SLNs - targeted lung cancer therapy. Angiopep-2 functionalized SLNs - antiglioma therapy. Sugar-grafted SLNs (galactose) - targeted liver cancer therapy. Conclusion: Nanoparticles with surface functionalization allow site-specific, safer, and more effective cancer therapy, but challenges remain in tumor penetration and large-scale production.

The Department of Pharmaceutical Sciences organized an insightful guest lecture on Nanocarrier Based Drug Delivery: Revolutionizing Targeted Therapy for B. Pharm. students on 23rd August 2025 (Saturday). The distinguished session featured Dr. Sistla Ramakrishna, Chief Scientist, CSIR-IICT, Hyderabad, as the eminent resource person. The program began with a cordial welcome address, followed by an introduction of the speaker, highlighting his notable academic achievements, research expertise, and significant contributions to the field of nanotechnology. The event was chaired by Dr. Ch. Jithendra, Head of the Department, and meticulously coordinated by Dr. M. Venkata Ramana, Assistant Professor, who ensured the smooth execution of the program. In his lecture, Dr. Ramakrishna emphasized the limitations of conventional cancer therapies such as surgery, chemotherapy, and radiation, which are often associated with non-specificity and severe side effects. He elaborated on the emerging role of nanotechnology in cancer therapy, focusing on Solid Lipid Nanoparticles (SLNs) as versatile carriers for both hydrophilic and hydrophobic drugs. A key highlight of the session was the discussion on surface functionalization with ligands (folic acid, transferrin, antibodies, peptides, sugars, etc.) to achieve targeted drug delivery and enhance therapeutic outcomes. The lecture also featured case studies, showcasing the applications of functionalized nanoparticles in various cancers-such as folate-grafted SLNs for improved brain delivery of docetaxel, adenosine-conjugated SLNs for breast and prostate cancer, transferrin-functionalized SLNs for lung cancer, and galactose-modified systems for liver cancer. These examples demonstrated advancements in bioavailability, toxicity reduction, and site-specific action. Concluding the session, Dr. Ramakrishna provided valuable insights into the future of nanomedicine, underlining the potential of multifunctional and personalized nanocarrier systems, along with the challenges of large-scale production and clinical translation. In his closing remarks, Dr. Ch. Jithendra expressed heartfelt gratitude to the guest speaker for his enriching presentation and motivated students to integrate the knowledge gained into their academic and professional pursuits.
